In this review, the DNA adducts obtained from four classes of carcinogens for which there is substantial evidence of human exposure are considered: N-nitrosamines, aflatoxins, aromatic amines, and polycyclic aromatic hydrocarbons. For each class, the metabolic activation pathways that lead to DNA adduct formation will be discussed briefly. The encoded protein is part of the NER (nucleotide excision repair) complext which is responsible for repair of UV radiation-induced photoproducts and DNA adducts induced by chemical carcinogens.
